yes, checkups wouldnt hurt. (tire pressure, all fluids, check wipers) you might want to get some fuel system cleaner to put in your tank (like techron)

On all my trips, I didnt check anything. I think the only thing I did once was to make sure the tires are properly inflated for mileage reasons.
Like the other guys said, smoking a cig, open window, music, it only helps for like 5 mintues. Also, I dont know what your schedule is like, but Thanksgiving weekend is the worst traffic day in America. I suggest beating Chicago Rush hour. There is a huge difference between stressful and relaxed driving.

I guess I am not the only one on a 12 hour road trip. I'm leaving Wednesday morning as well. Leaving from Los Angeles, Ca to Rock Springs, Wyoming for Thanksgiving with some relatives.
Things I do are checking the tire pressure, brakes, fluids, a good nights sleep, some coffee and tea, a good breakfast, and some nice songs where I won't get tired of listening too.

Day before:
Eat well - If you don't eat well and then have to be in the car for 12 hours you WILL get sleepy. I'm not saying to go out and eat perfect, just don't end up having small stuff the day before
Sleep well - Once again, driving while sleepy sucks ass and is super dangerous. Get your rest, you'll need it.
Tires - Your link to the road. Super easy to check, and it's the most likely thing to cause you problems
Fill up - Duh
Clean windows - When you're driving around town, dirty windows aren't too big of a deal. But when you are on the highway for 12+ hours with little cover from the sun (which can sometimes end up and HORRIBLE angles), dirty windows will not only piss you off, but they will slow you down. While you are filling up, just clean them off. Having a small towel in the car with you can also help in case you have any fog issues or if handprints are causing grief.
Check Fluids - Don't go crazy but check what you have easy access to. Oil, Anti-freeze, windshield wiper, ect.
Pack - Don't wait until the day of the trip to pack. You'll forget something and having to load up all that stuff will push back your departure time and make you frustrated, leading to a crappy trip.
The Trip:
Water bottle - don't drink too much or you will stop too often. Take a sip every 15 minutes or so. This also helps pass the time
Apple/Banana - Good for you, easy to eat, and no real clean up. Plus, they will give you a good source of long term energy (Something that energy drinks/pills wouldn't be able to do). Don't even fell bad about tossing the core/peel out the window either. Just don't hit someone with it lol
Tunes - driving without music long distances suuuuuucks

Sugar/energy drinks are a ripoff....they only work in limited time....its proven.....
You need carbs for long term energy.
Check tires and fluids, just top everything off.
music, water, and snacks are a great.
Good sleep the night before and the most important thing to remember, is-------------------------------------------- STOP WHEN YOU NEED TOO!!!
Dont force it, dont ignore it. Know your limits.....if you feel even the slightest bit tired just pull off and smoke a cigg or stretch nap whatever it beats falling asleep dont be a hero or a cock just pull off and waste the time its not that serious, but your safety and the other drivers safety is serious!
I cant stress that enough, i use to drive 6 hrs one way from Hinesville Ga to Ft Lauderdale Fl every other month to go home from the Army....so i did that drive like 10 times....got some tickets but i never had 1 close call because i knew my limits and i listened to my body.
